La primer etapa comienza donde terminó el prólogo, en Rotterdam, cruzando por segunda vez el Puente de Erasmo  (Erasmusbrug).

Desde Rotterdam, la etapa apunta hacia el sur, para un breve recorrido por las costas holandesas, las mismas que fueron barridas por el viento durante la tercera etapa del Giro de Italia este año. Esta primera etapa del Tour se dirige hacia el interior, donde antes el Giro continuaba  por la costa, hacia  Middleburg, por lo que los vientos no afectarán demaisado esta vez. La carrera se dirigirá hacia Flandes, hogar de uno de los monumentos del ciclismo, el Tour de Flandes (Tour of Flanders). Tom Boonen ha dicho que el recorrido interior hacia Brusela será exigente y emocionante, y que esta etapa será importante para los corredores belgas.

Despues de una etapa llana de 224 km., hay un pequeño repecho poco antes del final, en cercanías del Estadio Rey Balduino (King Baudouin stadium) in Bruselas, para celebrar el cumpleaños 65 de  Eddy Merckx. Merckx, considerado el mas grande ciclista de todos los tiempo, nació en el este de Bruselas, en una pequeña ciudad llamada  Meensel-Kiezegem, el 17 de Junio de 1945, justo un mes despues del fin de la Segunda Gran Guerra. LA etapa pasará a través de  Meise, 10 km antes del final, donde Merckx actualmente vive.
